Thiobacillus ferrooxidans is an important leaching microorganism, which is acidophilic and suitable for medium temperature environment. It widely exists in acidic mine water and acidic environment containing iron or sulfur. In recent years, it has been widely used in biological desulfurization technology. So what should we pay attention to in the cultivation of Thiobacillus ferrooxidans?
據(jù)研究表明：1.氧化亞鐵硫桿菌適合在初始pH為2.0時(shí)生長(zhǎng), 此條件下, 細(xì)菌很快適應(yīng)環(huán)境, 可以正常吸收營(yíng)養(yǎng)物質(zhì), 合成新的原生質(zhì), 形成一定量細(xì)菌所必需的酶；
According to the research, 1. Thiobacillus ferrooxidans is suitable to grow at the initial pH of 2.0. Under this condition, the bacteria quickly adapt to the environment, can normally absorb nutrients, synthesize new protoplasts, and form a certain amount of enzymes necessary for bacteria.
2.氧化亞鐵硫桿菌的佳培養(yǎng)溫度是30℃.在這兩個(gè)溫度范圍內(nèi), 酶活性高, 細(xì)胞的生物化學(xué)反應(yīng)速度和生長(zhǎng)速度較快; 但微生物機(jī)體的重要組成, 如核酸、蛋白質(zhì)等對(duì)溫度較敏感, 隨著溫度的升高可遭受不可逆的破壞.
2. The optimum culture temperature of Thiobacillus ferrooxidans is 30 C. Within these two temperature ranges, the enzyme activity is high, the biochemical reaction rate and growth rate of cells are fast; but the important components of microbial organisms, such as nucleic acid and protein, are sensitive to temperature and can be irreversibly destroyed with the increase of temperature.
3.當(dāng)接種適量增大時(shí), 培養(yǎng)基中的初始菌數(shù)增加, 氧化亞鐵硫桿菌快速繁殖.但由于培養(yǎng)基中的營(yíng)養(yǎng)物質(zhì)有限, 加入過(guò)多的菌液影響細(xì)菌的生長(zhǎng)繁殖.表明培養(yǎng)氧化亞鐵硫桿菌的佳接種量為10%.
3. When the inoculation amount increased, the initial number of bacteria in the culture medium increased, and Thiobacillus ferrooxidans multiplied rapidly. However, due to the limited nutrients in the culture medium, excessive bacterial liquid was added to affect the growth and reproduction of bacteria. It showed that the optimal inoculation amount of Thiobacillus ferrooxidans was 10%.
